Greene County is located in the western part of the state of Alabama and was established on December 13, 1819. The county was initially part of the Mississippi Territory, and it later became part of the Alabama Territory created in 1817. Greene County is named in honor of General Nathanael Greene, a Revolutionary War hero who played a significant role in leading the American forces during the Battle of Guilford Court House in North Carolina.

Greene County has a rich history with various Native American tribes, such as the Choctaw and Cherokee, living in the area before European settlement. The county's fertile soil attracted settlers from various parts of the country, transforming the region into a thriving agricultural community. Cotton, corn, and other crops were grown in the area, and the Tombigbee River served as a transportation route for trading goods.

During the 19th century, the county played an essential role in the development of the Black Belt region of Alabama, known for its abundant fertile soil and large slave-holding plantations. After the abolition of slavery in 1865, tenant farming and sharecropping became the backbone of the county's economy.
