The county's fertile land led to thriving farming communities that were supported by the transportation networks formed by the Ohio River, and later by railroads and highways.

The city of Greenup, recognized as the county seat, was initially named as the small but thriving city of Greenupsburg. It was founded in 1803, alongside the establishment of the Greenup County. The name change to Greenup occurred in 1872.

Iron production was a significant industry in Greenup County during the later part of the 19th century, with several iron furnace facilities operating in the area. However, this industry gradually declined after the introduction of cheaper steel production methods.
