Marlboro County is located in the northeastern part of South Carolina and was established on March 2, 1785, as one of the state's seven original districts. The county derived its name either from the Duke of Marlborough or natural deposits of marl found in the region. With the city of Bennettsville as its county seat, Marlboro County played a vital role in South Carolina's early history.

It was initially inhabited by the Native American tribes called the Saura, Waccamaw, and the Pee Dee. Eventually, European settlers, mostly of Scottish, English, and Welsh origins, started to migrate to the area in the late 17th and early 18th centuries. The economy during this time relied heavily on the agricultural sector, specifically on the production of cotton, wheat, and corn.

Throughout its history, Marlboro County was the site of some significant war-related events. The Battle of the Great Cane Break was fought in 1775 during the Revolutionary War, while the Civil War saw several Sherman's troops marching through Bennettsville in 1865.

In the 20th century, the county's population grew and expanded its economy beyond agriculture, with manufacturing and retail industries playing significant roles. Notable residents, such as singer-songwriter Laurin M. Shaw, who wrote "It's a Long Way to Tipperary," also contributed to the county's character and history.
