Established in 1834, it was named after the Des Moines River, which in turn derives its name from the French phrase "la Rivire des Moines," meaning "River of the Monks." The county originally included a significant portion of the current state of Iowa, as well as parts of Minnesota and Dakota Territory, but was later divided into multiple smaller counties.
The county seat of Des Moines County is Burlington, which played an important role in the history of the region. Founded in 1833, it was a significant center for steamboat transportation and an economic hub during the mid-1800s. Burlington also served as the capital of the then-Iowa Territory from 1838 to 1841, before the capital was moved to Iowa City.

Constructed in 1894, and originally intended to connect the downtown area to the residential neighborhood, Snake Alley has been dubbed the "Crookedest Street in the World." With a steep incline and five half-curves and two quarter-curves, it has attracted tourists from around the world, who come to marvel at its unique design and winding nature.
